Follow your institution’s SOP and biosafety rules. This article describes common research-lab practice; it is not a substitute for hands-on training.
Before you open the vial
- Verify the batch on tararesearch.ie/verify before breaking seal — match batch code and COA.
- Work at a clean bench or biosafety cabinet with gloves and eye protection.
- Allow a cold-shipped vial to reach room temperature in the bag if condensation is present — reduces moisture shock to lyophilised material.
- Have BAC water, alcohol swabs, and your reconstitution worksheet (calculator) ready.
Opening the vial
- Remove flip-off cap if present; do not touch the rubber stopper.
- Wipe stopper with 70% isopropyl and allow to dry fully (~10 seconds).
- Use a sterile syringe and needle (or vented needle) sized for your BAC volume.
Adding BAC water — critical technique
Direct high-pressure stream onto the powder can denature the compound, create foaming, and yield inconsistent concentration. Always introduce diluent gently down the inside wall of the vial.
- Draw the planned BAC volume (the calculator defaults to 2.0 mL — adjust only with a documented reason).
- Insert needle through the centre of the stopper at a slight angle so the tip rests against the glass, not the powder.
- Inject slowly down the side of the vial, letting the liquid run down the glass onto the cake.
- Remove needle; do not shake. Roll gently between palms or swirl until the solution is clear — may take 1–3 minutes.
- If material sticks to the stopper, a very gentle swirl with the vial inverted once is acceptable — never vigorous shaking.
Label the vial immediately: compound, concentration, reconstitution date, and batch/verify ID. Store reconstituted material refrigerated per your SOP.
