This article explains syringe graduation maths for laboratory aliquots. It is not guidance on injection technique, sites, or human use.

A U-100 insulin syringe is graduated so that 100 units equal 1.0 mL. Therefore 10 units = 0.10 mL, and 1 unit = 0.01 mL. The reconstitution calculator outputs draw volume in these units once concentration and target mcg are known from your approved protocol.

  1. Confirm vial concentration (mcg/mL) after reconstitution — calculator or notebook.
  2. Convert protocol target mcg → mL: volume_mL = target_mcg ÷ concentration_mcg_per_mL.
  3. Convert mL → U-100 units: units = volume_mL × 100.
  4. Read the mark that matches that unit count; never estimate between poorly lit marks.
Tip

Keep a printed conversion strip (0.1 mL = 10 U) taped near the balance / hood for double-checks.
